龙源期刊网 http://www.qikan.com.cn 肠炎清对 ICUC 大鼠结肠组织 TLR4、 NF—κB 蛋白表达、TLR4mRNA 的影响 作者:黄亦彤 钟志勇 吕永慧 蓝天美 潘竞锵 武昕 来源:《云南中医中药杂志》2016 年第 09 期 摘要:目的通过观察肠炎清对免疫复合溃疡型结肠炎模型(ICUC)大鼠结肠组织 Toll 样 受体 4(TLR 4)和核因子-κB(NF-κB)表达的影响,探讨肠炎清治疗 IBD 的作用机制。方法 采用三硝基苯磺酸(TNBS)加免疫复合法造模。将大鼠随机分为空白对照组、模型对照组、 肠炎清低、中、高剂量组(833、1667、3333mL/kg)、柳氮磺吡啶(SASP)组(05g/kg)。 造模后第 2 d,用药组分别给予相应药物灌胃 7d,给药 7 天后麻醉处死大鼠,取新鲜结肠标 本,采用免疫组化法检测 TLR 4、NF-κBp65 的表达,实时定量 PCR 法(qRT-PCR)检测 TLR 4mRNA 的表达。结果免疫组化结果:NF-κBp65 和 TLR4 在各组各只动物均可见阳性表达,其 中肠炎清高剂量组及 SASP 组可显著下调 NF-κBp65 的表达,有统计学差异(P005)。PCR 结 果:与模型对照组相比,各给药组大鼠结肠组织 TLR4 表达均为阳性,但未见统计学差异 (P>005)。结论下调 TLR4、NF-κBp65 的表达,是肠炎清治疗 IBD 的作用机制之一。 关键词:肠炎清;溃疡性结肠炎;ICUC 大鼠;TLR4;NF-κBp65 中图分类号:R2855 文献标志码:A 文章编号:1007-2349(2016)09-0072-04 【Abstract】Objective: To observe the effect of Changyanqing on the colon tissue Toll-like receptor 4 (TLR 4) and nuclear factorκB (NF-κB) expression of immune complexes ulcerative colitis (ICUC) rats and explore its mechanism to treat IBD. Methods: Trinitrobenzene sulfonic acid (TNBS) plus immune complex was used to make models. Rats were randomly divided into a control group, a model control group, Changyanqing groups with low, medium and high dose (833, 1667, 3333mL/kg) and sulfasalazine (SASP) group (0.5g/kg). In the following the day after modeling, the treatment groups were administered with corresponding drugs for 7 days, and the rats were sacrificed 7 days later to take their fresh colonic samples. Immunohistochemistry was used to detect TLR 4 and NF-κBp65 expression, and RT-PCR method to detect TLR 4mRNA expression. Results: Immunohistochemical results: NF-κBp65 and TLR4 in each group can be seen positive expression, among which the expression of NF-κBp65 of Changyanqing group with high dose and SASP group could be significantly down-regulated, and there are significant differences (P005). PCR Results, compared with the model group, TLR4 expressions of the colon tissue of each administration group were positive, but there was no significant difference (P>005). Conclusion: Changyanqing can down-regulate TLR4, NF-κBp65 expression, and its mechanism may be related to the regulation of TLR4 / NF-κB signal pathway. 【Key words】Changyanqing, ulcerative colitis, ICUC rats, TLR4, NF-κBp65 龙源期刊网 http://www.qikan.com.cn 属非特异性的自发免疫性疾病,病变主要累及结肠黏膜和黏膜下层,临床主要表现为反复 腹痛、腹泻、黏液血便等。其病因和发病机理至今仍不清楚,既往研究认为,细胞因子网络的 失衡在 IBD 的发生发展中占有极其重要的地位。*年来不少研究发现,炎症介质及细胞因子 可导致机体多条信号通路活化,直接或间接地影响炎症介质的表达,导致肠黏膜受损产生。 肠炎清是由我院自行研制的纯中药制剂,临床用于口服和灌肠治疗 IBD、慢性腹泻、结肠 息肉电切术后致肠黏膜受损、肠道菌群失调及肠功能*取8靡┲
